    摘要:

    当归是一种大宗中药材,其变异株的挖掘与发现,对当归种质资源创新与功能基因组研究具有重要的意义。为分析当归矮化株系表型与植物生长抑制对成药期当归生长发育的影响,用植物生长抑制剂矮壮素(CCC)和多效唑(PP333)溶液对成药期正常当归植株进行叶面喷施,观察植物生长抑制剂对当归表型及早薹和成药生长的影响,分析当归自然矮化株系及生长抑制剂处理引起的当归矮化株表型与矮化生长的机理,揭示赤霉素在当归早薹中的作用,探讨利用植物生长抑制剂降低当归早薹率和提高药用根产量的可行性。结果表明:自然生长条件下可出现当归矮化变异株型,正常当归植株的CCC和PP333溶液喷施,不仅导致早薹当归薹茎等生殖器官结构伸长功能的丧失,还引起未早薹营养生长当归的株形矮化,但对早薹率无显著影响(P>0.05)。这一结果得到植物春化机理与发育转变方向的表观遗传学调控理论支持。最终得到结论,大田成药生长当归的早薹可能与赤霉素开花调控途径无关,旨在减缓当归早薹的CCC和PP333喷施没有显著作用。

    Abstract:

    Angelica sinensis (Oliv.) Diels is a major traditional Chinese medicinal herb, and the exploration of its mutant strains is of great significance for the innovation of A. sinensis germplasm resources and functional genomics research. To analyze the dwarf phenotype of naturally occurring dwarf strains and the effects of plant growth inhibitors (PGIs) on the growth and development normally growing of A. sinensis at the medicinal maturation stage. The foliar sprays of chlormequat (CCC) and paclobutrazol (PP333) (two common PGIs) were applied to normal A. sinensis plants at the medicinal maturation stage. The effects of PGIs on plant phenotype, early bolting and growth at the medicinal maturation stage were observed. The mechanisms underlying the dwarf phenotype and dwarfing growth induced by natural mutation and PGI treatment were analyzed, the role of gibberellins (GAs) in early bolting of A. sinensis was clarified, and the feasibility of using PGIs to reduce the early bolting rate and increase the yield of medicinal roots was discussed. Naturally occurring dwarf mutant strains were found in A. sinensis under natural growth conditions. Foliar application of CCC and PP333 solutions to normal A. sinensis plants not only led to the loss of elongation ability of reproductive organs (e.g., bolting stems) in early-bolting plants, but also caused dwarfism in non-early-bolting plants at the vegetative growth stage, with no significant effect on the early bolting rate (P>0.05). This result was supported by the epigenetic regulation theory related to plant vernalization mechanism and the direction of developmental transition. The early bolting of field-grown A. sinensis at the medicinal maturation stage may be independent of the GA-mediated flowering regulation pathway, and foliar application of CCC and PP333 has no significant effect on alleviating early bolting. This study revealed the phenotypic variation characteristics of A. sinensis dwarf mutants and the effect of PGIs on early bolting, which lays a foundation for the exploration and utilization of high-quality mutant germplasm resources, and provides important guidance for the rational application of PGIs in A. sinensis cultivation.
