    摘要:

    [目的]准确测定青枯雷尔氏菌的菌液浓度。 [方法]选用胰蛋白胨大豆肉汤培养基和生理盐水为稀释液稀释青枯雷 尔氏菌 RsA(烟草分离源)和 RsB(番茄分离源)菌悬液,利用血球计数板计数法测定菌悬液浓度,并利用分光光度计法测定菌 悬液在 600 nm 处的吸光度值,建立青枯雷尔氏菌菌悬液浓度与吸光度值间的线性关系。 [结果]线性回归方程表明:胰蛋白 胨大豆肉汤培养基和生理盐水作为稀释液测定菌悬液浓度结果差异无统计学意义(P>0.05),但 RsA 和 RsB 菌悬液的吸光度 值具有差异,回归方程分别为 y = 6.578 4x+0.158 28(r2 = 0.998 99)和 y = 5.162 7x-0.125 82( r2 = 0.998 99),回归方程间斜率和 截距均差异有统计学意义(P<0.01)。 [结论]建立的青枯雷尔氏菌的菌液浓度标准曲线可用于测定吸光度值后快速计算不同 分离源青枯雷尔氏菌菌悬液浓度,且无需将菌悬液培养基置换为生理盐水进行吸光度测定,具有一定的便捷性和实用性。

    Abstract:

    [Objective]Accurately determine the concentration of Ralstonia solanacearum. [Method] Tryptone soy broth medium and physiological saline were used as diluents to dilute Ralstonia solanacearum RsA( tobacco source) and RsB (tobacco source).The concentration of bacterial suspension was measured by hemocytometer method,and the absorbance value of bacterial suspension at 600 nm was measured by spectrophotometer.The linear relationship between the concentration of Ralstonia solanacearum and the absorbance value was established.[Result]Linear regression equation demonstrated that there was no significant difference in slopes between two diluents.However,the absorbance values of RsA and RsB bacterial suspensions were significantly different,and the regression equations were y = 6.578 4x+0.158 28(r2 = 0.998 99) and y= 5.162 7x-0.125 82(r2=0.998 99) respectively.[Conclusion]The concentration standard curve of Ralstonia solanacearum established in this study can be used to determine the absorbance value and quickly calculate the concentration of Ralstonia solanacearum suspension from different sources without replacing the bacterial suspension medium with normal saline for absorbance determination.The method reported in the study was provided with convenience and practicality.
