    摘要:

    对唾液链球菌的固体和液体培养条件进行优化,通过单因素试验和参考已有文献资料设计了不同的培养条件,优化得到 了唾液链球菌固体培养最优的条件是:MRS培养基,pH 7.2,温度37 ℃下在体积分数为5%的CO2的培养箱中培养。液体培养 条件是:按照乳糖7 g,蔗糖3.5 g,葡萄糖3.5 g,胰蛋白胨14 g,大豆蛋白胨7 g,酵母提取物7 g,磷酸氢二钾/磷酸二氢钾0.02 mol/L,硫酸镁 200 mg/L,硫酸锰 100 mg/L,氯化钙 60 mg/L,VitB1 10 mg/L,蒸馏水 1L 的比例配制液体培养基,pH 7.2,温度 37 ℃下在体积分数为5%的CO2的培养箱中静置培养。进行放大培养后,最终得到的冻干好的菌粉共计10.9 g,活菌数达到 4.9×1011 cfu/g。为工业生产提供了有效的信息。

    Abstract:

    The solid and liquid culture conditions of Streptococcus salivarius were optimized by single factor test and referring to existing literature. The optimum conditions for solid culture of Streptococcus salivarius were obtained as follows: MRS medium, pH7.2, temperature 37 ℃ in incubator containing 5% CO2. The optimum conditions for liquid culture of Streptococcus salivarius were obtained as follows: according to the ratio 7 g of lactose, 3.5 g of sucrose,3.5 g of glucose,14 g of tryptone,7 g of soybean peptone,7 g of yeast extract,0.02 mol/L of potassium dihydrogen phosphate,200 mg/L of magnesium sulfate,100 mg/L of manganese sulfate,60 mg/L of calcium chloride, 10 mg/L of VitB1, distilled water 1L to prepare the liquid culture medium, pH 7.2, temperature 37℃ in incubator containing 5% CO2. After enlarged culture, the final lyophilized bacterial powder was 10.9 g, and the viable bacterial count was 4.9×1011cfu/g. It provides effective information for industrial production.
