    摘要:

    目的 几丁质酶属于糖苷水解酶家族,在植物生长和胁迫反应中起关键作用。本研究旨在通过全基因组鉴定高粱几丁质酶基因家族,解析其基因组特征与进化规律,并结合公开转录组数据与自主开展的定量反转录PCR(quantitative reverse transcriptase-mediated PCR, qRT-PCR)试验,分析该基因家族在蚜虫胁迫下的表达响应特征,填补高粱几丁质酶基因家族响应高粱蚜胁迫的研究空白,为高粱抗蚜分子育种挖掘候选基因。方法 通过生物信息学方法鉴定了42个高粱几丁质基因(SbChi),系统分析其理化性质、染色体定位、基因结构、进化特征及顺式作用元件;结合RNA-seq数据与qRT-PCR试验验证SbChi基因家族在蚜虫胁迫下的表达模式。结果 这42个SbChi基因家族成员包含GH-18和GH-19这2个结构域,5个亚家族,其不均匀分布在高粱的9条染色体上,其中有3对基因发生片段复制,分化时间分别约4.06、4.08和8.15百万年;SbChi蛋白多为酸性蛋白。顺式元件预测揭示了许多胁迫响应元件的存在,主要涉及激素响应、生长发育调控等,表明该基因家族主要参与胁迫抵抗。使用已发表的RNA-seq 数据,发现了SbChi基因家族在抗性和易感高粱品种之间的差异表达。qRT-PCR分析证实了蚜虫胁迫下SbChi基因家族成员的相对表达特性。结论 研究结果为阐明SbChi基因家族在生物与非生物胁迫响应中的潜在功能提供了新见解,并为解析高粱适应胁迫环境的分子机制奠定了理论基础。

    Abstract:

    [Obiective]Chitinases belong to the glycoside hydrolase family and play a crucial role in plant growth and stress responses by hydrolyzing chitin. This study aims to genome-widely identify the chitinase gene family in sorghum, analyze its genomic characteristics and evolutionary patterns, and investigate the expression profiles of this gene family under aphid stress by integrating public transcriptomic data and independently performed quantitative reverse transcriptase-mediated PCR(qRT-PCR) experiments. This work will fill the research gap regarding the response of the sorghum chitinase gene family to sugarcane aphid (Melanaphis sacchari) stress, and provide candidate genes for molecular breeding of aphid-resistant sorghum.Method In this study,42 sorghum chitinase genes (SbChi) were identified using bioinformatics approaches. Their physicochemical properties,chromosomal locations, gene structures, evolutionary characteristics, and cis-acting elements were systematically analyzed. The expression patterns of the SbChi gene family under aphid stress were further validated by integrating RNA-seq data and qRT-PCR experiments.Result These 42 SbChi gene family members contain two domains (GH-18 and GH-19) and are classified into 5 subfamilies, which are unevenly distributed on 9 chromosomes of sorghum. Among them, 3 pairs of genes underwent segmental duplication, with divergence times of approximately 40.60, 40.84, and 81.49 million years ago, respectively. Most SbChi proteins are acidic proteins. Prediction of cis-acting elements reveals the presence of numerous stress-responsive elements, mainly involved in hormone responses and growth and development regulation, indicating that this gene family is primarily involved in stress resistance. With published RNA-seq data, differential expressions of the SbChi gene family between resistant and susceptible sorghum varieties have been found. qRT-PCR analysis confirms the relative expression characteristics of SbChi gene family members under aphid stress.Conclusion The results of this study provide new insights into clarifying the potential functions of the SbChi gene family in biotic and abiotic stress responses, and lay a theoretical foundation for analyzing the molecular mechanism of sorghum adaptation to stressful environments.
