    摘要:

    种子纯度是衡量种子质量的关键指标,种子纯度的鉴定也是黄瓜杂交种商业化育种过程中至关重要的一步。‘川翠13号’黄瓜是四川省农业科学院园艺研究所育成的华北型黄瓜新品种,为了构建‘川翠13号’种子纯度鉴定体系,快速、简便、准确地鉴定杂交种子的纯度,选取均匀分布在黄瓜基因组上的240对SSR分子标记,从中筛选出符合亲本间呈互补带型的简单重复序列(simple sequence repeat, SSR)标记,并通过田间形态鉴定与掺假法验证SSR标记鉴定黄瓜种子纯度的准确性。结果表明:(1)从240对SSR标记中共筛选到2对符合种子纯度鉴定标准的SSR标记(Cs007和Cs087),SSR标记鉴定结果与田间形态特征鉴定结果完全一致;(2)掺假法验证结果说明,筛选到的2对SSR标记为‘川翠13号’种子纯度鉴定的特异标记,其余由本课题选育的黄瓜品种无法使用。本研究构建了以特异性SSR分子标记Cs007和Cs087为核心引物的‘川翠13号’杂交种子纯度快速鉴定的技术方法,大大缩短了杂交种的纯度鉴定周期,提高了鉴定效率,为该品种的商业化育种提供了技术支撑。

    Abstract:

    Seed purity is a key index to measure the quality of seeds, and the identification of seed purity is also a crucial step in the commercial breeding process of cucumber hybrid. ‘Chuancui13’ is a new cucumber cultivar bred by cucumber research group of Horticultural Research Institute, Sichuan Academy of Agricultural Sciences. To identify the purity of hybrid seeds quickly, simply and accurately, the simple sequence repeat (SSR) markers showing complementary bands pattern between the parents were screened out in 240 SSR molecular markers. The accuracy of SSR markers was verified by field morphological identification. The results show that :(1)two pairs of SSR markers (Cs007 and Cs087) show polymorphism differences between two parents; (2)the accuracy verification results are consistent with the field morphological identification results. The results show that the two pairs of SSR markers (Cs007 and Cs087) are the specific markers to identify seed purity identification of ‘Chuancui13’. A technical method of the purity identification of ‘Chuancui13’ hybrid seeds is constructed using specific SSR markers Cs007 and Cs087 as the core primers. It has greatly shortened the time of hybrid purity identification, improved identification efficiency, and provided technical support for the commercialization breeding in cucumber.
