    摘要:

    目的 采用湿法消解-荧光光度法测定87份不同来源苦荞种子的硒含量,以初步确定硒含量较高的苦荞资源,进一步选育富硒苦荞品种。方法 首先以硒的添加回收率和消解时间为评价指标,选择样品消解温度,然后采用湿法消解-荧光光度法测定87份苦荞种子资源的硒含量,初选富硒苦荞品种。结果 4个消解温度下,硒的添加回收率为87.2%~102.4%;消解温度越高,消解时间越短,但温度过高,样品容易爆沸和局部蒸干,使回收率降低,综合考虑,消解温度选择240℃。10个苦荞种子样品硒含量的标准偏差为0.45。87份苦荞种子的硒含量变化幅度为 0.0 109~0.1 790 mg/kg,平均值为0.0 560mg/kg。结论 优选出含量较高(> 0.0 500 mg/kg)的苦荞品种42种,为富硒苦荞产品的研究提供了理论依据。

    Abstract:

    Objective The selenium content of 87 Tartary buckwheat seeds from different sources was determined by wet digestion and fluorescence spectrophotometry in order to identify the resources of Tartary Buckwheat with high selenium content and further select and cultivate the seed rich in selenium.Methods The selenium content of 87 Tartary buckwheat seed resources was determined by wet digestion-fluorescence spectrophotometry with the recovery rate and digestion time as the evaluation indexes.Results At four digestion temperatures, the recovery of selenium was 87.2%-102.4%. The higher the digestion temperature is, the shorter the digestion time; whereas the higher the temperature is, the easier the sample is to explode and evaporate locally, which reduces the recovery rate. Considering all factors, the digestion temperature is selected at 240℃. The standard deviation of selenium content in 10 Tartary buckwheat seed samples was 0.45 by wet digestion and fluorescence spectrophotometry. The selenium content of 87 Tartary buckwheat seeds varied from 0.0109mg/kg to 0.1790 mg/kg, with an average of 0.0560mg/kg.Conclusion 42 varieties of Tartary buckwheat with higher content (> 0.0500 mg/kg) were selected, which provided a theoretical basis for the study of selenium-rich Tartary buckwheat products.
